Kidding aside, run 2-3 anti spywares.
I normally run Lava soft and AVG antispyware.
There is no "best" as such. Among the worst I would stay away from norton as it itself is a spyware.

I am already going linux mint most probably.

For windows, I would recommend widows firewall at the strictest level along with lavasoft adware. Relax options in the firewall according to your usage. Works well for me.

For the total peace of mind, just do these.

1. Boot up computer with live Linux CD/DVD (Ubuntu/Koppix/SuSE/Mandriva etc.)
2. Do not mount your Windows hard drive in Linux (unless required while net surfing).
3. Attach a USB drive to save downloaded files/Linux settings while surfing.
4. Connect to internet (via cable modem or ADSL)
5. Turn on Linux firewall (if available - no worries if not either)
6. Surf as much you want - you Windows hard drive is protected from virus/spyware!
7. After surfing complete, reboot your computer with Windows normally.

However, if you are using windows software modem (aka winmodem via dial up connection), the above method may not work as Linux will struggle to set up your net connection.

Combination of s/w

From my painful experience I have found that a combination of Anti-Spy softwares together with a good antivirus software is best.

I'm using Spybot and Ad-Ware with Yahoo Antispy (This is surprisingly useful sometimes!). To complete the loop I have Kaspersky Anti-Virus. I had Norton, found it to be USELESS!!! McAfee is better than Norton, but Kaspersky has protected me from persistant trojans and spyware that the others (including Spybot and Ad-Ware) could only detect but not remove.
